Accurate values for vmax and Km can be determined from the Lineweaver-Burk plot. The Michaelis-Menten equation can easily be converted to 1/v0 = (Km/vmax x 1/[S]) + 1/vmax which is analogous to the formula for a straight line, y=mx+b. If 1/v0 is set to 0, then 1/vmax is the y-intercept and would be 0.01 umol/min and -1/Km would be the x-intercept and would be calculated to be 0.13 mM.
