The initial velocity for each trial was plotted against the concentration of lactose. Note that 6 more trials were plotted and that the concentration of lactose increases by 0.5mM for each trial. The concentration of lactase remains constant for each trial. From this curve, the maximum velocity of the reaction, vmax, can be estimated. This is the point at which the curve reaches a plateau. Vmax may be estimated to be 75 umol/min. The Michaelis-Menten constant, Km, can also be estimated. This is the point on the curve that is half way up the curve before it starts to plateau. Km may be estimated to be approximately 0.12mM. In order to get accurate values for vmax and Km, a Lineweaver-Burk plot can be made.
